Ipamorelin

is a synthetic pentapeptide that acts as a highly selective growth hormone secretagogue (GHS), meaning it stimulates the pituitary gland to release natural growth hormone (GH). It mimics the action of the hunger hormone ghrelin but is noted for a "clean" pharmacological profile, as it does not significantly elevate levels of cortisol or prolactin.

Mechanism of Action

Ipamorelin binds to the ghrelin receptor (GHSR) in the hypothalamus and pituitary gland. This action triggers a natural, pulsatile release of GH into the bloodstream, which is considered more physiological than exogenous (synthetic) GH injections. This results in increased levels of Insulin-like Growth Factor 1 (IGF-1), which mediates many of the beneficial effects.

Potential Benefits

Benefits are primarily linked to the resulting increase in GH and IGF-1 levels:

  • Muscle Growth & Recovery: Supports protein synthesis, accelerates tissue repair, and reduces recovery time from injury or strenuous exercise.
  • Fat Loss: Enhances the breakdown of fat stores (lipolysis) and improves metabolism while preserving lean muscle mass.
  • Improved Sleep: Helps facilitate deeper, more restorative slow-wave sleep stages crucial for recovery and overall health.
  • Bone and Joint Health: Promotes bone mineralization and collagen synthesis, potentially reducing joint and muscle pain and improving bone density.

Safety and Regulatory Status

  • FDA Status: Ipamorelin is not an FDA-approved prescription drug for general use. It was investigated in phase II clinical trials for the treatment of postoperative ileus (gut paralysis) but was discontinued due to lack of efficacy. The FDA has also raised safety concerns, flagging it as a substance that may present risks, including immunogenicity or adverse reactions when administered intravenously.
  • WADA Status: Ipamorelin is a prohibited substance at all times under the World Anti-Doping Agency (WADA) Prohibited List.
  • Availability: It is commonly marketed as a "research peptide" for laboratory use only and is not intended for human consumption outside of a medical setting.
